Taking total RNA from Hela cells as template, the B2M (A) and GUSB (B) gene were amplified using HiScript II One Step qRT-PCR SYBR Green Kit (Vazyme, #Q221) and a similar kit of another brand, respectively. The Q221 demonstrates a higher amplification efficiency, indicated by a lower C_t value under the same template input and less primer dimers (almost none).

The Vazyme HiScript II One Step qRT-PCR SYBR Green Kits are designed for SYBR Green I-based qPCRs that directly use RNA as templates. The reverse transcription and PCR can be finished in one tube, significantly reducing pipetting procedures and the risk of contamination. The HiScript II Reverse Trancriptase and Champagne Taq DNA Polymerase contained in this kit and an optimized buffer system enables high-sensitive total RNA detection (as little as 1 pg).

The HiScript II One Step qRT-PCR SYBR Green Kit is a master mix system. The 2× One Step SYBR Green Mix contains an optimized buffer, dNTPs, specificity-enhancer factors, and SYBR Green I. The One Step SYBR Green Enzyme Mix contains HiScript II Reverse Transcriptase, RNase inhibitors, and Champagne Taq DNA ploymerase.

b. contains HiScript II Reverse Transcriptase, RNase inhibitor, and Champagne Taq DNA Polymerase.

c. Used to rectify the error of fluorescence signals between different wells. Use 50× ROX Reference Dye 1 for ABI 7900HT/ 7300 Real-Time PCR and System and StepOne PlusTM；Use 50× ROX Reference Dye 2 for ABI 7500, 7500 Fast Real-Time PCR System and Stratagene Mx3000P. Don’t use ROX for neither Roche nor Bio-Rad Real-Time PCR instruments.
